Der-depletion alters ribosome stability and/or biogenesis, as evidenced by polysome profiles (Bharat, 2006). YihI stimulates Der GTPase, possibly acting as a negative regulator of 50S ribosome assembly and retarding early exponential growth (Hwang, 2010). Der has two tandem GTP-binding domains. der is an essential gene. Der and YihI form a complex, as does Der-delta200 (lacking the N-terminal 200 aa domain of Der) and YihI-delta45 (lacking the N-terminal 45 amino acids of YihI) (Kobayashi, 2009; Hwang, 2010). Der and YihA are paralogs, with YihA matching the C-terminal GTPase domain better than the N-terminal GTPase domain. The YhiI stimulation of Der GTPase activity requires both Der Ser16 and Ser216, indicating both Der GTP-binding domain activities are required for Der GTPase activity (Hwang, 2010).
